AmVac Initiates Phase III Trial with its Lead Vaccine Gynevac for the Treatment of Bacterial Vaginosis

ZUG, SwitzerlandJune 18, 2014 /PRNewswire/ — Swiss-based AmVac AG, a private biotech company focused on the development of innovative vaccines, announced today the initiation of a Phase III study with AmVac’s lead vaccine Gynevac in Bacterial Vaginosis (BV). The trial aims to confirm the safety and efficacy of a new generation of the vaccine produced in accordance to the current GMP standards. The previous version, containing Thiomersal, had been approved in Hungary in 1997 for the treatment of certain gynecological inflammations and has since then been applied successfully to over 200,000 patients suffering from various urogenital conditions with no significant side effects recorded.

The placebo-controlled, double-blind, randomized study will enroll 240 patients. The primary endpoint is the treatment of BV confirmed by the Nugent scoring system. In addition, the trial will monitor changes in vaginal mucosa and evaluate several immunological markers. Five clinical centers in Hungary will participate in the trial. AmVac expects to include the first patient in June 2014 and to complete patient enrollment within about nine months.

About bacterial vaginosis (BV)

BV is among the most common conditions found in women health care settings with an estimated 47 million patients in the five major EU markets, the US and Japan. The disease occurs as the result of disbalance in vaginal ecosystem. Typical symptoms include abnormal discharge, burning, itching and discomfort. BV is particularly critical because it is also a door opener to serious secondary infections, such as HIV and other sexually transmitted diseases (STD). Further, the association of BV with preterm birth is confirmed by numerous studies. The current standard therapy is antibiotic treatment, which is associated with undesired side effects, e.g. development of antibiotic resistance or allergic reactions. Moreover, the protective vaginal flora often does not recover completely. As a consequence, disease recurrence rates after antibiotic treatment is high. Clinical studies with the previous version of Gynevac showed dramatic improvements in terms of efficacy, safety and long-term protection.

US Android Users on Average Engage 138 Minutes Daily with their Smartphones

BOSTON, June 13, 2014 /PRNewswire/ — According to Strategy Analytics’ AppOptix platform, US Android users have been spending more time with their devices. Android smartphone users on average spent 138 minutes with their device in May 2014, an increase of 20% from the usage in December 2013.

The analysis is based on over one million individual application sessions on more than 1500 Android smartphone users in the US during the period. Neurim Pharmaceuticals Announces Publication of Positive Effects of Add-on Circadin(R) in Alzheimer’s Disease Patients

TEL-AVIV, IsraelJune 19, 2014 /PRNewswire/ — Neurim Pharmaceuticals announced today publication of the results from an exploratory Phase 2 randomized placebo-controlled clinical trial evaluating the safety and efficacy of add-on Circadin® (Prolonged Release melatonin 2mg) to standard therapy in Alzheimer’s disease (AD) patients. The study, published in the Clinical Interventions in Aging Journal, demonstrates positive effects of the drug on cognitive performance and sleep maintenance in the AD patients.

“Endogenous melatonin levels are reduced already at preclinical AD stages. Because melatonin is important for good sleep quality and because poor sleep quality has recently been linked to AD, it was important to investigate whether replenishing the missing hormone would be beneficial in AD patients and whether such effects would be related to the improvement in sleep,” said Dr Tali Nir, head of clinical trials at Neurim Pharmaceuticals.

In this study, 80 patients diagnosed with mild-to-moderate Alzheimer’s disease, with and without insomnia co-morbidity, receiving standard therapy (acetylcholinesterase inhibitors with or without memantine) were randomly assigned in a double-blind manner to 2 mg of Circadin® or placebo treatment nightly for 24-weeks. The paper reports that patients treated with Circadin® for 6 months had significantly better cognitive performance than those with placebo as measured by Instrumental Activities of Daily Living (IADL) and Mini Mental State Examination (MMSE). Mean Alzheimer’s Disease Assessment Scale – cognition (ADAS-Cog) did not differ between groups. Sleep efficiency as measured by Pittsburgh Sleep Quality Index (PSQI) Component 4 also improved with Circadin®. In a subgroup of patients suffering from comorbid insomnia, Circadin® treatment resulted in significant and clinically meaningful effects vs. placebo in mean IADL (p=0.032), MMSE (+1.5 vs. -3 points, p=0.0177) sleep efficiency (p=0.04), and median ADAS-Cog values (-3.5 vs. +3 points, p=0.045) . The treatment was well tolerated.

This publication comes at an exciting time when the causal relationship between sleep disturbance and the AD-relevant accumulation of beta amyloid in brain was discovered,” said Prof. Zisapel, CSO of Neurim Pharmaceuticals. “This study demonstrates the significance of good sleep quality and melatoninergic mechanisms in improving both cognition and sleep problems in Alzheimer patients and calls for further focus of this mechanism in AD treatment.”
